Which Alexa dye do people prefer for use with a 543 laser, Alexa 546 or 555? Thanks!

Alexa 555 has a pretty low quantum yield.  I generally prefer 546,
though I've never used either with a 543 laser.

We use alexa 555 standard with the 543 laser and it’s great—fairly bright and stable (for fixed sample labeling at least).  I would hesitate to use 546 with the 543 laser…having a look at the Invitrogen fluorescence spectra viewer (no commercial interest) it will show that 543 laser will excite alexa 546 at about 40% efficiency, but alexa 555 at about 70% efficiency….of course these measurements are done in particular buffers etc. etc…just something to consider…

We initially used Alexa546, but have since switched to Al555, as the 546 seemed to bleach rather quickly in our hands. We find the 555 more photostable.
A number of groups here also use Cy3 with good results.
