Based on the spectral viewer the 568 will obviously be excited better but I've never used it so I don't know how it compares with regards to quantum efficiency and photostability etc? Other dyes that we frequently use with this laser are Cy3, Rhodamine-B and TRITC. http://listserv.acsu.buffalo.edu/cgi-bin/wa?S1=confocal Adrian, both dyes can be excited with the 561nm laser. Alexa 568 is often used here in our facility - without bigger problems. If you are looking for a dye especially for this laser, the 568 gets my recommendation. 594 also works, but cannot be used in combination with something excited with 633/640 (Alexa 633/647, Draq5 etc.).
