An ideal airy diffraction pattern has about 4/5 of the power in the main airy disk, the rest being in the rings. In the real world though scattering will will drastically decrease the light that gets through the pinhole. https://spie.org/publications/fg01_p88_airy_disk Craig On Wed, Jun 15, 2016 at 2:44 PM, Kai Schleicher <[hidden email]> wrote: > ***** > To join, leave or search the confocal microscopy listserv, go to: > http://lists.umn.edu/cgi-bin/wa?A0=confocalmicroscopy > Post images on http://www.imgur.com and include the link in your posting. > ***** > > Hi, > > Does anyone have a reference to clarify how much light in % is rejected by > a confocal pinhole at 1 AU compared to a widefield microscope? > > Thanks and cheers, > Kai > > -- > > Kai Schleicher, PhD | Research Associate in Advanced Light Microscopy | > Biozentrum, University of Basel | Klingelbergstrasse 50/70 | CH-4056 Basel | > Phone: +41 61 267 22 50 | [hidden email] | > www.biozentrum.unibas.ch | www.microscopynetwork.unibas.ch > |

So if your sample is 3D (from the microscope's point of view, that is, much thicker than the z-resolution of the microscope), which is most of the time, the pinhole will reject *most* of the light (99% in a thick densely labeled specimen), when compared to widefield... The Airy disc figures (> 80% transmission) are only meaningful for thin samples (thin layer, subresolution beads, etc), if you want to compare to widefield.
