Cholesterol dyes

Search the CONFOCAL archive at
http://listserv.acsu.buffalo.edu/cgi-bin/wa?S1=confocal

Hello all!!

I have a user who is wanting to stain his cholesterol in some muscle tissue for
a fixed slide.  He already has filipin which works alright in a fluorescence
microscope, but the problem with our imaging is that we are not equipped with
the correct laser for excitation.

So basically I was wondering if anyone knows of a green, red, or far red dye
that can be used to stain cholesterol in fixed muscle tissue.

To my knowledge there are 3 things that you can use

Laurdan

di 4 ANNEPSHQ for membrane fluidity - less fluidity more sterols -
ratiometric

cholera toxin CT-B - several Alexas

I must point out that filipin III is actually the worst since is
extremely toxic and and bleaches very fast.

Regards,
NM

PS: I have no commercial interest but they can be purchased from
Molecular Probes/invitrogen
