Consumables for grant application

Hi

I'm putting together a grant application to establish a small, but I hope, effective Imaging Facility. We are asking for a Leica SP2, that we already have, to be ‘recomissioned’ (hasn’t been serviced for several years) and to be upgraded to MP capability, a Deltavision core system, laser dissection, two widefield systems and a manager’s salary for 3 yrs. From talking with others it would appear that most users of such facilities bring their own consumables but I guess it would be useful to ask for some running costs on the grant application. I have no real idea of what internal consumables such a facility might require and/or how much to ask for per year. Could someone suggest some appropriate consumables and costs?

Hi Colin,

Interestingly, and to my surprise, objectives can be considered consumable.  I had to have our 63x Zeiss objective reworked (~$2800)after having oil leak into it, and Zeiss was not interested in covering it under the service agreement.  Other than that, Hg lamps, immersion medium, lens cleaner, cotton swabs, lens tissue, lab tissues and a couple of commercial slides for testing and calibration are the expenses for the multiple system I oversee.  Maybe a stash of slides and coverslips for those who come umprepared might be useful also.  As you say, most users supply their own consumables.

Hi Colin,
Carl covered the majority of the consumable costs.  Some software may  
have service contracts, as well, to cover major updates and tech  
support.  I second the need for a budget to cover repairs to lenses,  
maybe scope cleaning.  Don't overlook  tools (screw drivers, forceps,  
wrenches, etc.). voltmeter, light meter, maybe some additional  
immersion oils of varying RI, some small lamps for work lights and a  
vacuum cleaner with a HEPA filter.  A small budget for upgrades to  
scopes and computers, if you can get away with it.   A spindle of CD-R  
and DVD-R may assist those who forget their disks.  Are you using  
nitrogen bottles for the air tables or installing a system for cleaned  
house air?

Dear List,

I would greatly appreciate if you could share your experience working with
the orange-red fluorescent proteins with the excitation max from >560 nm.

I have noticed that both mCherry and mStrawberry fusion proteins have
tendency to aggregate in association with membranes, and are clearly more
cytotoxic than CeFP, YFP (or venusYFP), tested as fusions with a relatively
large numbers of proteins (>10), predominantly membrane proteins.

Would you know: which protein among mRFP1 derivatives (or other REDs) is the
most soluble in vitro/in vivo?

Hi Colin,
The only thing I'd add to what Carl and Glen have covered is a service
contract or extended warranty on the confocal.  We've had a full extended
warranty on our SP2 since purchase in 2000, and it has covered replacement
of two lasers, a motherboard in the scanhead, motorized stage, several
x-galvos (when they went through a flakey stage), other minor glitches  and
annual maintenance visits.  The systems may be more stable now, but we think
this was well worth it.
