Hi,

my question is about the standart approach to STORM. Using a Cy3-Cy5 switch
and appropriate activation and read-out lasers one is generally interested
in the emission of Cy5 as this is your super resolution signal.

Can one use the emission of the Cy3 (which I think should be present when
activating the switches) to get an ordinary "low-res" image of the sample?

Or do the switches work differently and there no emission in the Cy3 channel
during activation? I'm aware that this signal will be minute as the
activation intensity is very low. But assuming one would turn up the
activation intensity (activationg to many switches)? Do you think there will
be noticable emission in the Cy3 channel, or even the Cy5 channel (FRET?)

Thanks,
Christoph
