Hi everyone,

we have an Olympus 25x water dipping objective and would like to run 5 day or longer time lapse imaging experiments.

After a few days imaging in growth media we clearly see an infection (probably bacterial) with the objective dipped in. A dish next to it, without the objective does not show an infection.
We tried cleaning the objective with EtOH (basically dipping the tip in it) and Virusolve (gentle wipe) and that still does not help.

Does anyone (maybe even someone from Olympus ???) have some tips how to disinfect or sterilise the objective without harming it?

Thanks,
Martin

I was just going to propose Cavicide1... but it's almost the same as
Virusolve.
It's ethanol, isopropanol, water and some quaternary ammonium salt. It's not
corrosive...
I would not be afraid to dip the lens into cavicide for couple minutes and
wash with ethanol and DI water (but make sure not to bring any contaminants
from the objective barrel down to the front lens during washing). No wipes
or tissue.
But check with Olympus first.
Best, zdenek

Zdenek,
Dipping alone will not remove the crud.  Use an cotton applicator stick wrapped in lens paper with your lens cleaner.  It is useful to do this under a dissection microscope.  Be extremely careful of using solvents.

good luck,
