FPS inquiry

Dear All
Can any one help us in providing the information on maximum frame rate per second in spectral mode achievable by point scanning LSM with atleast 4Kx4k pixel resolution  and 16 bit digitization at 512x512 pixels ?

Hi Ashok,
 
Can you give us some more detail? What do you mean by spectral mode (ie lambda scan or just a single emission capture)? Also I am unsure what you mean by 4kx4k resolution at 512x512. And finally what system (Nikon A1, Leica SP5, Olympus FV1000, Zeiss 710 Meta etc)?

Following are the specifications. Is it possible have system which can meet following specification by any of the manufacturer (preferably Zeiss or Nikon) ? 

1. Maximum scan resolution should be ≥ 4Kx4K pixels for all channels and should be selected freely down to 4x4 pixels.  2. System should be capable of acquiring minimum 5 frames per second @ 512x512 pixel resolution . Digitization capability of 8/12/16 bit should be available with the system in spectral mode.

Dear Ashok,
 
Both Zeiss and Nikon have fast scan confocal systems (5 Live and A1R respectively). They will gove you much hgher than 5 frames a second but acheive it in different ways. All modern confocal systems will capture 16bit data (but may only be using 12 or 14bit PMTs and promote the data to a 16 bit range). As to the range of resolutions availible both systems should be adjustable (i have limited experience with either so i am not sure, but there will be people on the list that will be able to give you much more accurate information).
 
In regards to spectral aquisition both Zeis and Nikon use a meta type detector, 32 small PMTs capturing the selected spectrum, as opposed to Leica or Olympus that move a single "gate" along. The advantage of the Zeiss and Nikon systems is they are much faster at collected a spectral or lambda series.
