Hi all,

Before reinventing the wheel, I was wondering if anyone has prepared a poll
to find out what equipment should be on one's list of future purchases.
There is so much going on now in light microscopy, and so many things from
multiple solutions for lightsheet, through FLIM, automated clearing
systems, 2 and 3p, brightfield systems, re-scan systems, mass cytometry
(not exactly light), simple live systems and more.

Thanks,
Avi

That definitely helps, but I have found that a lot more people say they are interested in a new technology than end up using it.  There's an army of beautiful, rarely used instruments out there.  For example I remember seeing a multiphoton head that spent its whole working life as a DAPI laser.  I endorse finding how many people are already committed to the kind of work where X technology will clearly help, along with new faculty who have hands-on experience with the tech in question, and giving weight to their needs.

Agreed! A fluorescence widefield system with a good camera is adequate for
most work. Throw phase and/or DIC in there and you have almost all the
bases covered.

I agree with Tim about PI's expressing keen interest in the latest technology but then never showing up to use it. The other key problem in a core is competing with individuals or departments that have similar instruments like a confocal. I think it is important to have a really well equipped basic confocal (e.g., White light laser, bunch of HyD (or equivalent detectors), environmental box) so that you have an edge over the lab that has a low end "personal" confocal that could siphon off your user base.

You also need a wide-field system to attract the labs that don't do any microscopy so you can groom them into becoming serious microscopists using confocal or whatever. This needs a really good camera attached to it.

After that, get people to commit a little money to bigger upgrades like STED or MP so you can see the depth of their interest.

Good points Thomas! Instruments at a higher level of sophistication are
best relegated to core facilities: The cost of the instruments is high
enough that a single researcher can often not justify the purchase.
Additionally, the core facility can maintain the on-site expertise
necessary to operate the equipment long-term. I have often seen high-end
equipment sitting unused as the one postdoc that knew how to use it has
long since left. The staff of a core facility maintains the knowledge base
for the equipment.

While I have seen exactly what Timothy describes below, I find this is mostly the case when an individual lab sequesters a piece of equipment.

For a shared resource, I have found that there is a ramp up time between a new technology and users.  Microscopy non-experts need time to understand what new technologies can do for them and plan experiments accordingly.

This is the model whereby you identify a useful technology, build or buy it, advertise it, and they will come.  But it takes time after the instrument is available before you develop a user base.  Whereas emails, fliers, technology seminars, etc are necessary to get out the word,  the best way to build this base is to have high visibility researcher(s) explaining in their scientific talks and informal meetings how the new technology addresses key research issues.

The survey route for identifying technologies may yield information, but my experience has been that they are ineffective.   On the other hand, talking to researchers directly about their projects and thinking about how microscopy technologies you have heard about could address their research is a great way to identify need.

Cheers-

Michael Cammer, Sr Research Scientist, DART Microscopy Laboratory

Hi, I agree with both Tim and Michael, however I think the "if you build it they will come" model may not be fruitful. Although I do agree that as the resident microscopy expert you are better suited to choose equipment than the PIs at your institution. Too many times I purchased items that were never used early in my career. There is a big divide between PIs stating that they want something and actual using it (this can be related to things outside your sphere of influence such as cost of materials or personnel time, etc). Even if the PIs are willing to write a letter of support for a purchase is not a solid indicator of usage.
I like to think of what I would need to write a successful NIH S10 Equipment Grant; you would have to show a need so great that several funded PIs are willing to drive to other institutions to use similar equipment or to ship samples around the world to achieve their results. In other words, preliminary data verifying the need and expertise to use the technology effectively.

It is a complex question that does not have a straightforward answer.

    Thanks,

    Avi
