How to upgrade an old fluorescence microscope to LED fluorescence microscope?

Hi,

The Hg lamp transformer on our Leitz DMRBE microscope (manufactured in the early 90's) died recently. Since, getting replacement parts for this "old, but so nicely functioning" microscope becomes more and more difficult we were wondering whether it is possible to upgrade this system to a LED fluorescence microscope.
The optics are in good shape, all of the dimensional stages worked just fine but the UV light source causes headache. The microscope is not used for live cell imaging but for screening tissue samples for autoimmune reactions to certain neuronal proteins.

It would be great if  I could get advice on what is needed to turn that microscope into a LED fluorescence microscope.
Any advice, shared experience would be very helpful.

Hi Manja,

One of our previous workers, Sergei, did something similar with our FV1000.
I never used a DMRBE, but it is probably a similar idea.

"1" is the coupler from the LED light source (it came as part of the
package - the light guide goes into the back side and is locked in by the
silver knob seen in the picture).
This coupler fits into "3", which leads to the microscope via another fiber
(or directly into the microscope body).

The old part ( http://imgur.com/Wyrztf9 ) used to connect to "3".
But as the new coupler is slightly smaller than it, we had to have a ring
made "2",  that allowed the new coupler to remain centered and be locked
in.
(Hint: The ring itself has a tiny screw in it to lock the ring itself to
the coupler, then the coupler+ring is locked in by the externally
accessible screw.)

This won't work of course, if your original is already narrower than the
new part, something I doubt.

Hi Manja,

consider ThorLabs LED4 (your choice of wavelengths), controller, liquid
light guide and LLG adapter.

Your microscope probably has old, degraded, Leica branded, filters. The
current generation hard coated filter sets should perform much better.
Simplest is to purchase one Quad cube and purchase ThorLabs LEDs that
match the exciter peaks of -- for example -- the Semrock
LED-DA/FI/TR/Cy5-A-000

which is what I have on my lab's DMI6000 microscope

You can use Semrock SearchLight or Chroma's equivalent spectral viewer,
to evaluate additional options.

Dear Maria,

you might want to check CoolLED (no commercial interest). We have
successfully used their products to upgrade several systems from Hg to
LED and are very happy with their solutions.

They have a choice of adaptors,

It may be better though to contact them for advice on your system.

Hi Manja,

I first of all must declare a commercial bias.

There should be no problem putting an LED excitation system onto your Leica scope.

One thing to be aware of is that most LED systems do not sufficiently cover the full
spectrum like the bulbs did. This should not be a problem if you can match up the LED peaks
with the excitation filter passbands that are in your microscope. If you are not sure just go
for an LED system like our CoolLED pE-4000 that has 16 peaks from 365-770nm.
