Mammalian cells need to be under saline all the time, so relative
humidity there is always 100%. Above the media, yes, needs to be 100% -
this would cause condensation on the lid, unless the lid is warmed a
little (if inverted epi-fluorescence only, the water on the lid does not
matter; evaporation of the media always matters).
Yes, 35 mm glass bottom dishes should be supported.
Unless you are going to be able to offer as good (or better) products
for a lot lower end user price, and as good or better service, than
