I used fluorescently labelled fibronectin and it did a very similar job. Although it was ages ago, I have a mosaic on my Andor system now not a FRAPPA. Essentially you need a fluorescent thing you can make a hole in and then calibrate the xy galvos of the FRAPPA to the position of the hole if my memory serves me correctly. I'm sure flurosecently labelled agarose would do the same job. The Fluorescently labelled Fibronectin can be brought from Thermo for about £350. We used, and still use, fluorescently labeled agarose. Also, Zazzle (others probably work) marker on a coverslip works well. We also use the Zazzle marker for TIRF demo. Dot the coverslip, and then add water. Some of the Zazze juice sticks, and the rest forms mini-micelles. Works great, easy, and cheap!
