MQAE Cl sensitive dye

We wish to use MQAE to sense Cl. The published spectral data for this dye
indicate that it excites in the near UV, and has a broad emission spectrum with
its peak in the mid 400 nm range.

We indeed see a diffuse blue fluorescence in cells loaded with MQAE,  when it is
excited with the DAPI cube (near UV excitation). However, we see a much stronger
green fluorescence when looking at it with a GFP cube (narrowband green
emission). The green emission is, in most cells, well localized to the nucleus.
We have of course looked at cells without MQAE, and in this case, neither the
blue nor the green emissions are observed. We also checked the cells in a
microplate reader and obtained consistent results.

The blue emission appears to be sensitive to Cl concentration, while the green
is apparently insensitive, but this is only and initial result from the
microplate reader, and we need to repeat it.

Can anyone on the list who is using or has used MQAE shed light on this matter.
