Measurements in Imaris

Hi

Have a facility user who is trying to make measurements using Imaris (version 7.3.1) and having some difficulties. Ideally she would like to use the slice section but cannot see how to do this in the software - maybe it is an upgrade we don’t have? Is there a way to do it in 3D consistently? When you move the navigator tool the line is not where you attempted to put it! Is there another software that is better to use?

Any help would be much appreciated!

Hi Ali

I’m not sure this is the same issue but we found that many functions in imaris move things to the nearest pixel so basically don’t work at sub pixel resolution.  Totally invalidates any particle tracking in imaris!

Dear Alison and Ian,

Thank you for using Imaris and I hope my reply below is helpful.

Alison, based on your description it sounds like the user needs to make a 3D line measurement.  This type of measurement is possible with our MeasurementPoints tool which can be used with our Ortho and Oblique Slicers.  You will find a short video tutorial covering this topic here: http://www.bitplane.com/learning/measurement-points-and-usage-of-ortho-and-oblique-slicers-video-tutorial   And please feel free to reach out to me directly if this doesn't solve the issue.

I also want to assure Ian and the entire community that spot detection in Imaris, including MeasurementPoints and Spots, places the center with subpixel precision.  When using automatic detection during the Spots wizard the precision depends upon the width of the applied filter (i.e. estimated XY diameter).

A possible place for confusion could be that the display of measurement values within Imaris is controlled by a preference for the number of visible significant digits.  This setting is adjustable and does not affect the export of these values to Excel, etc..

I'd like to ensure that we fully address your concerns so please reach out to our Support team to review images and analysis results where you've seen this spot placement behavior.
