Dear all,
We are interested in assessing the PSF of our microscope when imaging in Benzyl Alcohol/ Benzyl Benzoate (BABB) solution (used for tissue clearing). What do you use as a point source when the clearing medium involves such organic solvents?

In our hands, Polystyrene beads seem to be dissolved in BABB while Quantum dots have issues with aggregation.

Thank you in advance for any advice you may have,

Etai

 Hi Etai,

We have recently found that gold nanoparticles work well for PSF
measurement in solvents.  The scattering from them is extremely bright,
they do not photobleach, and a single nanoparticle can be imaged at various
wavelengths.
