Mounting media

Search the CONFOCAL archive at http://listserv.acsu.buffalo.edu/cgi-bin/wa?S1=confocal Following my question about mounting media for TIRF, I received an advice to use FluorSave from Calbiochem, and it works pretty well with the 100X, 1.46 Zeiss TIRF objective (RI is said to be around 1.38).  The sad thing is that Calbiochem just decided to stop producing Fluorsave, so I'm holding to the last bottle we have here !

For non-TIRF coverslips, I usually use Mowiol but I'm fed up with poor conservation (at 4°C, after one month the autofluorescence just ruins the signal) and lack of reliability (quality is very variable from one preparation to another). What mounting medium could replace Mowiol for fluorescently labeled, fixed, cultured cells? It would be better if it was hard-setting (or gel) and doesn't require sealing with nail polish or VALAP.

You may look into ProLong Gold from Invitrogen.
