My apologies

I apologize for my recent multiple mailing to the list. It took me a bit to catch-on to the fact that they were going to the list rather than just the person who the reply was to. Sorry about cluttering up everyone's mailbox.

Antibody labeling

Could someone recommend an IgG labeling kit? There are kits made by Invitrogen, Pierce, and others. Which in your experience is the best?

Thanks!

Mike

Re: Antibody labeling

I´ve used the alexa labeling kits from Invitrogen (alexa647 and alexa488) with good result. /fredrik

Re: Antibody labeling

To do staining with directly labeled antibody in one step. For our application it is important though to have all the fluorescent reagent bound to antibody, nothing should be left free in solution.

Mike,

Invitrogen and Pierce are both good companies but the former, as you
know, bought up Molecular Probes (MP) whose company focus was to make
end products for microscopists. Pierce was and I assume is still the
best supplier of reagents, columns, i.e., the raw materials for do it
yourselfers like me. Often I would use a reactive fluorophore
derivative from MP but dialysis cassettes, and small purification
columns from Pierce and also some specialty reagents such as the -SH
reducer TCEP, an excellent compound for revealing thiols for
dye-maleimide conjugation that does not need removal from the treated
protein before applying the maleimide as other disulfide reducers do.

I have in previous postings sung the praises of the Invitrogen Zenon
labeling kits, which use an Fab-Alexa dye (various colors) to bind
with a primary antibody before labeling cells. With the Zenon-labeled
primaries I successfully labeled cells on slides, and in cell
suspensions whether for confocal or flow cytometry.

Email me if you want more details. Both are great companies.
(Disclaimer: no financial interest on my part).
