http://listserv.acsu.buffalo.edu/cgi-bin/wa?S1=confocal Hi all, After several years break from the list I'm back on with an old question that is perpetually new. It is: What are people's recent multiphoton/confocal imaging experiences with new detectors like the hybrid photodetector (e.g. H8236 PMT/APD) from Hamamatsu or the new silicon PMTs (e.g. from SensL)? They seem to have nice fast responses and the gains are getting better (10^6 for the latter). Are PMTs still the popular choice? Related question: What bandwidth for PMT analog I-V conversion is required for fast scanning? Is 200KHz okay or does one need > 1MHz? Or, are you using photon counting instead of analog, these days? My two incentives for asking are: 1) I'm trying to keep my external detectors alive a bit longer on our ancient BioRad 1024MP and 2) I'm working on setting up another system. Thanks, Brian O'Rourke |

http://listserv.acsu.buffalo.edu/cgi-bin/wa?S1=confocal I've used the Leica APD detector (at the factory, I don't have one). Sensitivity is amazing but you can get frustrated since if the signal gets too strong it just cuts off and all goes black (rather than just saturated as happens on a conventional PMT). So you have to be careful setting up a Z-stack, and if there's a bright spot in your image when you want to look at dim structures you also have a problem.
