Dear microscopist’s community,
I’m not sure that it’s a correct forum, but there are a lot of good NIS Elements users here.
In our Nikon confocal system we have a digital colour camera also installed, as a incubation system, that’s why we can do wound healing experiments in BF.
The last user when he brought his multiwell plate for the acquisition, no any clean area was found to get a shading correction image by multiplicative mode. However, when the experiment was finished, we clean some area by a blue tip in one of the well, and with the same conditions of illumination made a Shading Image - _nooptconf_ acquisition. Now, do you now if it is possible to correct the experiment data with a posteriori made shading correction image. If this possible, could it be apllied to a mosaic application? In our case the option in the menu Image -> Background -> Substract Background Using Reference is in grey colour, I think it is not available. Is it any other form to make the correction?
