New instrument coming to the Advanced Imaging Center, Janelia

Dear Colleagues,

We are excited to announce that the Advanced Imaging Center (AIC) will be adding the SiMView light sheet microscope to our fleet for the upcoming round of proposal call, which will be announced next week.

The SiMView (Simultaneous Multi-View) light sheet system, developed by Philipp Keller, is engineered to have a large (800 X 800 um) FOV with lateral resolution of 406nm and Z resolution of 2 um. It features 5 laser excitation wavelengths spanning the optical spectrum for maximal fluorophore flexibility. The system is also equipped with an environmental chamber to ensure specimen health.  What distinguishes the SiMView system most prominently from commercial light sheet microscopes is Janelia's "autopilot" software, which can maintain correct image focus, light sheet orientation, and centering of ROI during the entirety of the experiment which could last for days. This unique functionality, via automatic feedback-controlled adjustment of up to 12 separate optical alignments, allows SiMView to accurately image the development of mouse embryos (or other similar size samples) for prolonged periods of time, even as the specimen grows in size and increases in refractive index heterogeneity.
