Hi

There was an email exchange about fluorescent spheres mounted on slides in an optical cement. I think it was about the prepared Molecular Probes slides. I've tried to search for the email exchange but can't find it. I'm trying to locate the supplier of the optical cement used which had an RI of around 1.5.

Does anybody remember the emails and the supplier?

Thanks

Colin

Hi,

I do not remember the email exchange but from what I learned at the Optical
Microscopy course at Woods Hole, Norland Optical Cement # 63 supplied by
Edmund Scientific should work. The RI for # 63 is 1.56.

Interestingly, I learned it from another Colin though it was Dr. Colin
Izzard!

Best regards,
Shiva

I think you're referring to an email I posted about a conversation I had with Iain Johnson at Molecular Probes about the TetraSpeck Beads (no commercial interest). I've copied it here again for you:

***********************
John:

The mounting medium is Saint Gobain Optical Cement.  According to my notes, this has a refractive index of 1.52 when cured.

The spectra for the TetraSpeck green dye are the same as our standard yellow/green FluoSpheres:http://www.invitrogen.com/site/us/en/home/support/Product-Technical-Resources/Product-Spectra.8811h2o.html

The spectra for the Tetraspeck dark red  dye are the same as our standard dark red FluoSpheres:http://www.invitrogen.com/site/us/en/home/support/Product-Technical-Resources/Product-Spectra.8807h2o.html

The blue and orange dyes are different from the standard FluoSpheres dyes so I don't have data for those at present.  I'll work on getting it but may take a few weeks.

Best Regards,

iain
