This question is about using PS-speck (Invitrogen cat# P7220) beads for the purpose other than they were designed for. I am interested in their refractive index. I have developed software for computing images under DIC of known specimens, and am considering these beads as test specimens. The goal is to compare computed images with experimental images.
I have tried polystyrene beads with known RI and small-size but they are hard to spot. I could spot PS-speck beads easily due to their fluorescence and then switch to DIC for imaging.
