Dear Melia
You might also consider CyGEL.
This live cell mountant will permit a gentle thermo-reversible immobilisation of your cells (live) and should further retain the 3-D structure since there is no adherence involved to create a "pseudopod" between cell and glass surface.

Cells are added to cooled CyGEL and deposited onto the slide where it will "set". Overlay the coverslip and re-chill to liquefy temporarily allowing the CyGEL to spread under the coverslip. Allow to warm above RT and it will re-gel.

Like this, cells should be good for 1.5-2h of imaging.

Standard formulation of CyGEL is PBS-based and a version ready for your own culture medium is also available - CyGEL Sustain.
