We are also in the dilemma of choosing laser of 140fs and 70fs which as the white paper suggest would have different fluorescent signal intensity and background. Our system contains an AOM an this contributes about 10,000 fs^2 to the overall GDD which is about 17000 fs^2. A 100 fs pulse would broaden to about 480 fs and a 70 fs pulse to 680 fs without pulse compensation.
