Re.:FRAP and TPE

*****
To join, leave or search the confocal microscopy listserv, go to:
http://lists.umn.edu/cgi-bin/wa?A0=confocalmicroscopy
*****

we did GFP in a membrane protein, check Certal et al. (2008) Plant Cell,
//*20*: 614-634

some more hints on :
Feijó, JA, Moreno N (2004) two-photon microscopy applications to plant
cells. Protoplasma, 223:1-32

pro's: greater zz' precision on the FRAPing
con's: as always, go easy on the throttle of the laser, it's a tiny
border between FRAP and fry... plus, globally speaking, in our hands it
was never as vital as normal laser confocal. Still, in our case, the
extra precision was useful.

g'd luck

There is a  paper by Thomas Oertner (Grunditz et all 2008 J. Neuro.) in
Switzerland in which the last figure is in vivo FRAP. They will probably be
able to help you,
Cheers,
Stéphane
