Hi, Eric,

Just to expand on what a couple other commenters have said...   For live cells, the calcein AM dyes are the best option for filling the cytoplasm in live cells.  Molecular Probes / Thermo Fisher sells green, blue, violet, and red-orange variants.  Be aware they will be pumped out of the cells over time and are not fixable.

For fixability after live cell labeling, I would recommend the CellTracker or CellTrace dyes.  These will label cytoplasmic proteins (though not as uniform of a cytoplasmic labeling as calcein dyes).

Other options exist for cells that are already-fixed, but I won't go into those here since that doesn't seem to be the focus.

All:

I am looking for a way to fluorescently counter-stain the volume of cultured cells, with a result similar to what would be achieved by having them express GFP in the cytoplasm. I have seen some references to staining with eosin Y. Does anyone have other suggestions?
