Hello,
The non-destructive immobilization of cells and objects for imaging can be
easily achieved using a patented, thermo-reversible hydrogel mountant
called CyGEL(TM).
In this thermo-reversible mountant, the sample can be recovered for
orthogonal analysis, be it culturing, sorting, DNA or RNA extraction, EM,
and so on.
In practice, CyGEL is cooled below RT to liquefy it and it can either be
mixed with or overlaid onto the cells/bacteria. A coverslip is then
placed on top.
The slide is cooled again to allow the gel to spread under the coverslip
and then wormed to set.
This should be good for a couple of hours without sealing the edges.
CyGEL is entirely compatible with fluorescence and can accept viability
dyes such as DRAQ7 to report dead cells.
