The (only) solution, which was recently offered to us by Leica, is to purchase a new workstation for ~9KEuro. And our team is not alone with this problem as I know at least 6 other microscopes with this issue. Did this group find an alternative solution to this problem? Did anybody successfully upgrade the OS on the existing hardware - without breaking the system? Second, with most (if not all) of our microscopes we're happier with Windows 7. It is unsupported, but it works; why break it then? When IT folks are complaining, my philosophy is, if a smart refrigerator, chinese watch, or a phone with Android 4 can connect to the institution's WiFi, why not a Win 7 PC? Third, if you don't get a more assuring answer from other listers, you can just "give it a shot": 1) Get a new harddrive (assuming your PC does not support NVMe I'd suggest a SATA SSD with good TBW specs, e.g. ADATA SU800, or, even better, Samsung 860 PRO (not EVO)). 2) now you have two options - upgrade the OS (higher chances of success?) or start from scratch (cleaner system?) 2a) To upgrade, clone your old disk to the SSD (it's a bit tricky if your old disk is bigger than the SSD). 3a) Boot from the SSD and do an in-place upgrade to Windows 10 (some versions are tricky to upgrade, e.g. Win 7 Enterprise). 4a) Test and report! or 2b) To start from scratch, install clean Windows 10 on the SSD (I remember some LAS versions could only run from the D drive, so if that's your case, and you want LAS to run from the same SSD, just partition it accordingly). 3b) Install the same LAS software you used under Windows 7 (the LAS may want to rewrite the firmware of the SP8, so if you use the same version of LAS, you'll be able to go back to your old OS later, if needed). 4b) Test and report! We can connect our microscope computers to our local server for data transfer, but they are cut off from everything else and will reject outside connections. It's not a perfect solution, but our IT department deemed it "good enough" given our needs and inability to upgrade them to Win10. This is to address our IT department encouraging elimination of Win 7 computers for network security. We found a second low-end computer and installed Win 10 Pro. Then we connected this directly to our Win 7 computer with a StarTech.com USB 3.0 to Gigabit Ethernet NIC Network Adapter. Our users have to transfer their acquired data from the Win 7 computer to the Win 10 computer at the end of their session (easy drag-and-drop). The Win 10 computer is accessible for remote data transfer. When I find some time we will dive into the approach of the LMU - integrating the dd (and avoiding the frimware update) as suggested by Zdenek. The short-term solution - at least for our systems - however, will be based on Kathy's suggestion: as our Microscopes already are directly connected to a Linux file server which is pulling the data automatically from the microscopes, we will simply take the SP8s offline and access the files exclusively through the server. Thank you for opening my eyes, there! I'll report back on the progress/success (much later) so others can profit from our experiences as I will from these of/at the LMU. Gratefully, aRnim PS: we are already running the ESU on these machines. So if your volumes have to be aligned in XYZ and have shear/mag issues, FIJI will require a lot of trial and error and could still result in slice to slice variation. Is this something you have to perform regularly or a one-off issue? If you want really effective N-D alignment I would recommend either going the hard route and coding it in Matlab or Python, or using a purchased software package like Imaris. If you want to keep to free software, BigWarp in FIJI can do a great job but is mainly main for 2D images. FIJIyama or the other built-in registration plugins are probably your best options. Typically these parameters are to be estimated from a known structure like a multicolored bead, and then applied to the biological image data acquired under the same conditions. Yes, "a bit of hacking around" describes it quite appropriately, I am afraid. Backup is essential, so that you can go back to the starting point, if everything else should fail. BTW, should you plan to upgrade to the linear scanning electronics (Klondike Upgrade), that would come with a Win10 upgrade included (and possibly new computer), since it does not run on Win7. From memory, it increases the duty cycle in unidirectional mode from 25% to 65% (i.e. the percentage of the total time during which photons are collected.) With the same line frequency and pixel size, pixel dwell time goes up. For some, it may be easier to convince funders to pay for 2.6x more photons in the same time period (or respectively shorter acquisition times), rather than a windows upgrade, although it is likely to be several times more expensive than what Arnim mentioned as Win10 upgrade costs. But I totally agree with others that fencing Win7 systems off the internet with a gateway computer is an efficient, fast and relatively inexpensive way to solve the problem in many settings. I did that once with a Win XP system hidden behind a NAS which had several Ethernet ports, one to the outside net, and one to the microscope computer. That worked very nicely. I am curious if you or anyone else who has gone the ESU route has had any issues with the updates interfering with the microscope control software at all? I know that sometimes security updates can include changes to the .net framework which I've been warned about in the past. Is this a valid concern? We contacted the university IT and we didn't even have to pay for it. The university had purchased limited extended licenses. Was a quick fix! |
