These membranes are mainly made of DOPC and similar phospholipid derivatives. The aim would be to get a positive control for a fluorescent molecule that is able to diffuse into the GUV and is accordingly detectable with Fluorescence Correlation Spectroscopy within the GUV. So, guessing from literature, I would need a dye that is neither too hydrophobic nor too lipophobic. Has anyone of you a dye in mind that is useful for my problem set? Help is much appreciated! Only if it diffuses inside it might also diffuse outside unless there the probe is charged (say, positive) and GUV has a negative potential. The acridine family is a good choice though, since they are 488 excitable, highly membrane permeable, and quite bright. If you have acridine orange or acriflavine around, I would try those first.
