Removal of Dapi

CONFOCALMICROSCOPY Digest - 20 Jun 2010 to 21 Jun 2010 (#2010-39)

Has anyone successfully removed not quenched, dapi once it has been bound to DNA?    It is interfering with downstream PCR.

Have a look at:

 

Multiparameter DNA flow-sorting demonstrates diploidy and SDHD wild-type gene retention in the sustentacular cell compartment of head and neck paragangliomas: chief cells are the only neoplastic component.

This paper describes the use of DRAQ5-stained cells, co-stained with anti-S100 antibodies flow-sorted for cell cycle position and S-100 status and then PCR’d to identify mutations (in disaggregated tumour tissue samples).
