I am trying to mount a PEN film on a glass coverslip and have it secure enough
to withstand the subsequent rigours of cell culture and immunofluorescence
processing. I can easily overlay the glass coverslip with the PEN film with
relatively little wrinkling of the film by floating the film on water or ethanol and
lifting underneath with the coverslip. It is specifically the securing of the PEN
film afterwards that I am having trouble with. I have tried coating the glass
coverslip with poly-L-lysine prior to mounting the film, and baking the
film/coverslip overlay overnight without success. I have also sealed the edges
of the film on the coverslip with nail polish but found that to be cytotoxic (I
assume something from the nail polish is leaching into the media). Does
anyone know of a way to secure the film to the glass coverslip, that would
allow for cell culture and immunofluorescence, or a non cytotoxic
sealant/adhesive that can be applied to the edges of the film after I mount it
on the coverslip? Any information is appreciated.
