TIRF CFP/YFP FRET cube for 457 laser line

Dear all
We have a Nikon Ti-E TIRF setup with an Melles griot argon laser. We are having trouble finding suitable FRET filter sets that fit the 457 nm side line, excitation is cut before 457. It is a fairly common laser as I understand it so there should be filters that match. Anyone else had this issue and solved it?

Hello Sebatian,
If your argon laser is about 40 or 50 mW, you may get enough power at 457 nm for TIRF. Argon laser peak intensity is at 488 nm, you can get enough power at 514 nm too, but, NOT at 457 nm.
We have the Melles griot argon laser (30 mW) with our TIRF. Of course we never used CFP-YFP pair. Currently we are using green diode laser and argon laser 488nm for TIRF FRET (GFP-RFP).

Are you talking about an excitation filter? You do not need any since this is a laser. Just buy a cube with dichroic mirror and emission filter.

Hi Sebastien,

If you contact me directly, I can help with an appropriate filter set,
mounted appropriately for TIRF.

BTW, yes you do also need an excitation filter. Lasers still need clean-up
filters, whether it's an Ar laser (with multiple lines which must be
completely blocked, not just by AOTF), or especially if any are diode
lasers.
