I have had difficulty tracking down a plasmid for TagRFP-T. I was wondering if anyone has this and would be willing to send me some? I could provide FedEx shipping information.

Someone in Canada would be preferred if possible.

405 laser for Leica SP5

Our 405nm laser on the SP5 has to be replaced (after only 2000 hours), I was
wondering if anyone has replaced this laser but got the laser directly from
Coherent and not through Leica representative. I was told by our
representative here in Israel that the lasers a custom made for the SP5 and
if I get one directly from Coherent they might not be able to align it in
the system. (the price quote for the laser through our representative is 4
times higher than the one I got from the Coherent representative)

Hi Claire,
UCSD is not in Canada, but Roger Tsien lists TagRFP-T  as a standard
construct that can be requested at    
http://www.tsienlab.ucsd.edu/Samples/Constructs.htm    -- see
http://www.tsienlab.ucsd.edu/Samples.htm  for instructions.

p.s. according to www.jcat.de  the TagRFP-T (Genbank EU582019) is not
mammalian codon optimized (also appear the case of many other FP's whose
sequences I've tried).

Hi Andre,

If this doesn't work, you can buy a cryo attachment from RMC that works
much better than any other attachment, in my opinion.  They seem to have
one for every microtome.  What's more, the RMC guys will even make custom
modifications to it if you have specific requirements.  Not a cheap way
out, but you'd never regret it.
