Greetings all,

I would like to purchase a temperature controller for our inverted microscope (IX-83) for use with high magnification oil immersion objectives. Ideally I would like to use a stage-only, peltier-based temperature controller for both heating and cooling (~10-40C). This is for sub-micron scale objects, so the heating/cooling unit must not generate vibrations which I would expect from fan-cooled units.

Does anyone have experience with this? Any recommendations for products?

Thanks,
Joe

Bioptechs has seen a significant trend toward micro-environmental systems that provide transitional temperature shifts for gene expression among other things.
The main factors other than temperature have been the speed at which the temperature changes and, as you pointed out, vibration.
The approach we have taken is to do the temperature transfer with two, external, thereby vibration isolated, Peltier controlled recirculating systems each at a different temperature.
We then set up a TTL activated solenoid switching system that provides the following conditions:
1. Temp A to the scope and back to itself while Temp B cycles back to itself
2. Temp B to the scope and back to itself while Temp A cycles back to itself
We have documented a 10 - 40 degree transition in less than 30 seconds excluding the objective, that takes a little longer.

On the scope we provide a fluid optical cavity such that the specimen on a coverslip has its own isolated space that is separated from the circulating temperature controlled fluid by a 0.5mm window.  If you are using high N.A. objectives, we use a temperature transfer collar on the objective.
This provides highly efficient heat transfer to the specimen and enables both reflected fluorescent and transmitted modes of microscopy.
For additional information contact Dan at Bioptechs. 724-282-7145.

Note, I would not recommend any type of system that relies on peripheral heat exchange due to the long equilibration times and thermal gradients that occur through peripheral heat propagation.

Dear Joe,

I have co-developed such a device 10 years ago, it is made available
through https://emblem.de/technologies/instruments/
Fast response mini stage temperature controller
We have used it to control temperature sensitive allels, it shifts between
