Dear community

I hope to have your help with our problem that maybe is a bit basic but we
would like to solve it with complete security.
We recently installed the technology of light sheet microscopy, SPIM (2
laser and 2 cameras), and with respect to calibration I have the following
doubts

   - What is the methodology to calculate the theoretical resolution axial
and lateral?, considering the scenario in which one rotates the sample and
   when one moves the savannah.
   - What is the recommended experimental methodology to quantify pixel
size?.  Consider that the samples should be mounted in capillaries.
   - Should we have any special consideration to obtain the experimental
PSF?, especially the type of movement in the z-axis.

Thank you very much everyone
regards
