Time-lapse in Slidebook

Dear All,

Has anyone done transmission time-lapse acquisition using Slidebook? Our problem is that transmission shutter never closes, and the sample remains exposed to light for many hours. It is white light, not fluorescence, but still.(Unfortunately, 3i has a policy of not talking to customers without a service agreement, so I have to seek help from the confocal community...).

Hi Michael,

(I assume your microscope transmitted light shutter works when toggled
manually ... can also think about investing in a transmitted light).

You could place a filter in the condenser light path -- ideally far red
or NIR -- and, if using fluorescence too, wavelength compatible with the
emission filter of whatever fluorescence cube is used for acquisition
just before or after the transmitted light image.

p.s. On our inverted microscopes, I usually cover the condenser opening
with a neutral density filter (and tape the edges so users don't do
anything to it), to both prevent dust from getting into the condenser
opening, and to attenuate the light of tungsten halogen lamp so that we
can use it at the max 12 Volts ... the only consistent intensity setting
(besides of course zero).

Good Morning Michael,

You are absolutely welcome to write an email to
[hidden email] and the team will certainly give their best
effort to help out with a quick software question (and true likely provide
an invitation to upgrade your older software to the newest version.)  What
you are looking for can also be found either by searching in the manual or
in the past 5 years or so, it can be found via pressing F1 and
context-sensitive help opens right up.

Here's the section of the manual describing how to do what you are looking
for:


   1.

   Setting Capture Preferences to Open and Close Shutter during Capture

If you are performing single channel 3D capture, the default fluorescence
shutter action in SlideBook is to leave the shutter open during capture. If
you would like the shutter to open and close after every capture, select
the Advanced button in the Capture Settings section of the Capture dialog
box. Select the 2D/3D capture checkbox labeled Open and close shutter on
single channel or simultaneous 3D capture in the General tab. The default
action for multiple channel capture is to open and close the shutter after
every capture.

For the approach you are taking, you can address this via this location in SlideBook properties:

properties > advanced capture > ShutterBrightfieldTimelapse

If you are capturing a single channel transmitted capture SlideBook leaves the shutter open, unless that’s set to yes. Therefore, if you'd like to shutter during single channel transmitted capture, please set this to yes. Please let us know if this resolves your issue.
