Hi All,

Did the various searches, but nothing seems particularly versatile.  What
we have seen is a fixed hole in the center of the top of a chamber, with
little to no play, even with the sealing gasket.

Does anybody have a favorite/s they could suggest?

We had a look into this last year (although haven't yet committed) and had
the Chamlide UM system suggested to us (no commercial interest):

We have used a Peltier stage from PhysiTemp with excellent success (TS-4SMP):

Since we do everything from 8 hour live cell imaging at 37oC to Drosophila pupae live imaging for 48 hours at 25oC to live  lamprey embryo imaging at 18oC, having a wide range of temperatures available is a must (this stage has a practical range of 0oC to 60oC, although in dry conditions it gets down to about -12oC).  Also, because it is a Peltier device, it ramps to the proper temperature and stabilizes in about 30s (think PCR machines).  We use tap water to remove waste heat, but they also have a water recirculator if you don't have access to a tap.

Also, the dish holder can be removed to mount slides, which we have used to keep live tissue sections at 4oC to prevent necrosis while imaging.

Originally we got the stage since we needed both heating and cooling, but we have found the temperature to be remarkably stable when measured at the sample (+/- 0.05oC), and we even use it for live imaging at room temp, as it will offset temperature fluctuations within the room, giving better consistency in our data.

For more advanced needs, the PID controller can also be reprogrammed.
