What we have seen is a fixed hole in the center of the top of a chamber, with little to no play, even with the sealing gasket. Does anybody have a favorite/s they could suggest? Thanks, as always. -- Best, Gary Laevsky, Ph.D. Confocal Imaging Facility Manager Dept. of Molecular Biology Washington Rd. Also, because it is a Peltier device, it ramps to the proper temperature and stabilizes in about 30s (think PCR machines). We use tap water to remove waste heat, but they also have a water recirculator if you don't have access to a tap. Also, the dish holder can be removed to mount slides, which we have used to keep live tissue sections at 4oC to prevent necrosis while imaging. Originally we got the stage since we needed both heating and cooling, but we have found the temperature to be remarkably stable when measured at the sample (+/- 0.05oC), and we even use it for live imaging at room temp, as it will offset temperature fluctuations within the room, giving better consistency in our data. For more advanced needs, the PID controller can also be reprogrammed. Hope this helps, Ben Smith Benjamin E.
