Abstract:

Based on the previous findings of Limoli and Ward, about 15 years ago Bonner
and colleagues used laser
microirradiation to induce DNA double strand breaks (DSBs) in cultured cells
pretreated with the nucleotide analog
BrdU (5’-bromo-2-deoxyuridine) as well as the DNA intercalating dye Hoechst,
following which the cells were fixed
for the analysis of DNA damage dependent phosphorylation of the histone
variant H2A.X. Since then many groups
have utilized these strategies and improved upon them to study the dynamics
of DNA repair proteins in living cells
