Hi there,
I would like to minimize my laser spot size during photobleaching using a Zeiss LSM 510 with a two photon tuned to 750nm. I was wondering if by using the "spot bleach" option does the scanning module raster across the image with the laser only exciting at the specified pixel, or do the scanning mirrors only orientate at a position corresponding to the specified pixel and then the laser excites? Will it reduced the effective photbleached area if the spot bleach was carried out the latter way i.e., by direct pointing of the scanning module and not by rastering?
