You draw a line over the 'wound' that cells are migrating to and the Kymograph provides a representation of how fast cells move along the line filling the wound. For non-linear cell spreading an area measurement is usually easiest, you have to threshold or process the image so the cell area is distinguished from background (generating a binary image is best for this). You can then display the growth of area covered by cells over time in Integrated Morphometry Analysis or using the Region Measurements tool. If the images are calibrated then measure the square microns of area, otherwise it might be best to measure a % coverage. MetaMorph opens several different formats, to find them all you can use the File - Open option and there is a drop-down at the bottom that lists Image Types. I would suggest the 16-bit tiff images whenever you are analyzing data, but when you save the images from AxioVision (or wherever you take your images) make sure there is no compression being used. Venkat, We use QuickTime Pro (US 29.99 for the Pro license last time I checked). With the Pro license, it is possible to import TIFF series or various movie formats, and convert them to Quick Time or other movie formats, with a variety of compressors, choice of frame rates, and image size.
