Search the CONFOCAL archive at
http://listserv.acsu.buffalo.edu/cgi-bin/wa?S1=confocalhi. we used a homemade closed perfusion system back in the day, and
measured subtle, quick changes in the cells (responding to pH
changes, etc.). it worked fairly well, you could really tell when
the new fluid came in, and there was only a tiny amount bathing the
cells, so changes were fairly quick and precise. on the downside, it
was difficult to get the cells into the chambers (in a leakproof
way). i haven't tested commercial chambers, but they should be
pretty good! the fluid flow was smooth, so that didn't affect
imaging, as far as we could tell. you could use valves to switch
between pre-gassed solutions.
