Dear friends;
I am using a Leica TCS-SP5 confocal microscope simulataneously with
elctrophysiology setup (Multiclamp 700B). To mount the electrodes we have
micromanipulators from Luigs and newmann (brand name 'Luigs neumann
junior'). For last 15 days we are getting constant one sided drift in the
recording elctrode that is about 5 micrometer within 5-10 min in z
direction , while 10-20 micrometer in y direction. We have beem trying
hard to rectify it but all in vain. I have tried following steps but
nothing worked out:
1. put off the manipulators and observed if yhe problem is electronic,since the
problem persisted, i concluded it is a mechanical problem
2. We tightened all the screws again and again but didn't worked, sometimes
depending upon the level of tightening and side of the screw from the pair (as
every attachment is attached with a pair of screws) the direction of the drift
reverts to opposite to the screw more tightened, but i never got a proper
balance, so i suspect there may be something else causing the problem.
3. We dismantelled the complete set and reassembeled again.
4. We have one set spare and i assembeled it also but that too showing the same
drift.
5. We tried to adjust thetension due to connecting cables by hanging them in a
direction such that they are in line with the electrode holder,at the same
angle.
6. We gave little lubrication to the parts recommended by manufacturer,
according to thier prescription.
7. Vibration free table is working fine.
Though the step nos. 2 and 5 show some improvement but the problem is not
cured completely, and i can not play much with the screws and i don't think
the very little cable tension will cause so much drift, as the instrument was
not that delicate earlier and it should not be rather.
If anyone uses this kind of set up and can suggest any solution i will
be greatly obliged.
