We are currently trying to figure out how to stimulate specific tissues or even single cells in zebrafish to induce the expression of a cre-recombinase that is under the control of a heat-shock promoter (hsp70). Since we have no experience in heat-shock induction by irradiation, I wanted to ask if anyone of you has ever done it and could give me advice. What kind of laser is suitable? Can it be done with near-infrared light? Is it possible to activate a heat shock promoter using a two-photon microscope? Will the zebrafish be damaged?
We are planning to mainly activate the hsp70-promoter in transparent zebrafish embryos. However, we also want to induce the promoter in adults. The Cre-recombinase expression would eventually lead to the expression of GFP in the heat-shocked cells/tissues.
I would be very thankful for any advice or help.
