mounting media

Dear all,
 
I would like to take your opinion about the mounting reagent that i should use for fixed cells in acceptor photobleaching experiments.
Which is the best mounting reagent for fixed cells in acceptor photobleaching experiments?

Hi Serra,
 
you can use any non-fluorescent media for mounting, except for anti-fade reagents which prevent the photobleaching of the acceptor dye. When we use formaldehyde for fixation at the end of the experiment, we leave it on the cells, otherwise, we use PBS or glycerol.
If you need an efficient software for the analysis of your images, I recommend our free ImageJ plugin:

I think any non-fluorescent media can be used except for antifade reagents which prevent the photobleaching of the acceptor. When we do formaldehyde fixation at the end of an experiment, we usually don't change it. Otherwise, we use PBS, glycerol or 1:1 PBS:glycerol.
