we have just purchased an SP5 confocal trying to do calcium measurements in smooth muscle cells.
Do anybody have any experience with calcium measuremenst with SP5.

Hi Pierre,

We have the same leica SP5 and we have done calcium imaging in the neurons with the help of perfusion system. if you can give some bit of detail about your experiment than i may suggest u some thing.

Hi Manish,

the last 20 years we have used dynamic video imaging to measure calcium in cultured human detrusor smooth muscle cells and smooth muscle cells from myometrium cultured in monolayers on 0.17 glas coverslips using fura-2 and indo. Now we would like to se investigate calcium sparks starting in ER near the caveolae.

Fura-2 WL is in the UV (340 nm). I need to know which dye is usable, which laser do I have to etc. Merely, a protocol. We would like to do perfusion but we do not know which perfusion chamber to use.

Anything will help me to move forward.

I would like to add that we succed to isolate "Cajal" cells (resemble nerve cells) from the urinary bladder and we would like to measure calcium in these cells too.
