optimal settings for photoactivation/readout PA-JF-646 dye

Hi all, wondered if anyone can help us. Last week we tried out some expts using the PA-JF-646 SNAP dye and we used the Zeiss Elyra system equipped with the 405 nm laser line for photactivation  of the dye and the 642 nm laser line for read out  of the single molecules.

From the recordings at the start of the expts we performed, it appears to me that the 405 activation laser was turned on for too long or there was too much laser power because I observed that lots of molecules were activated and a lot of excess signal was generated in the recorded image.

With regard to the PA-JF 646 dye, can anyone provide any information what settings are required for optimal photoconversion and readout of this dye is using the Zeiss Elyra system, regards John
