Hi Adrian,

We have one of the new Olympus 25x 1.05NA objective installed. In short
it is absolutely amazing. The image quality, s/n ratio etc. compared to
our older 20x 0.95NA is phenomenally better. We have reduced excitation
power by about 1/2 to 2/3 on our regular samples (zebrafish expressing
GFP) @920nm. The resolution while technically only slightly improved
looks a hell of a lot better. The correction collar is also a major
advantage as we can now compensate for the embedding media of out
samples some what.

I can answer some, but not all of your questions.

- Transmission of the objective is meant to be ~82% out to 1000nm, not
sure what it is at 1400nm but since out MaiTai doesn't tune that far I
haven't had to find out.
- The field number is 27.5. This is achieved by creating a "virtual"
back plane inside the microscope somewhere. The field is not any larger
than the 20x though due to the system being limited to 18mm FN (though
this could be higher in the current generation FV1000 MPE systems that
have an adaptable excitation path).
- Not sure about the overfill question, hopefully Olympus can chip in
with that answer.
- The thread is identical to the older 20x lens. So if you want a bigger
filed you can always swap them over (assuming you have the adaptable
light path, otherwise you will have reduced performance.)

Hi all,

Does anybody know what are the specs for the new Olympus 25x 1.05 N.A.
objective or the Nikon 25x 1.1 N.A. objective ?

More specifically:

- What is the transmission of the objective up to 1400 nm ?
- From what I know all Olympus objectives are designed to be used with a
180
mm tube lens. Is this also for this Olympus objective ? Then the
effective
focal length of the objective would be 7.2 mm.
- What is the Field Number of the objective ? Suppose this is 22 mm like
for
the 20x 1.0 NA, is it correct then that the diameter of the image plane
of
the sample for which the objective is designed is 22/25 mm=0.88 mm.
- What is the diameter of the back-aperture that has to be overfilled
with
the laser beam to get the best PSF ? I intend to overfill it with a
truncation ratio of 1.
- What is the location of the paraxial lens equivalent of the objective
within the objective ?
- Is the thread type of this objective the same as for the 20x 1.0 N.A.
?

Did anybody work with these objectives so far ? What is your experience
?
