I have a copy of Lakowicz's "Principles of Fluorescence Spectroscopy"
and find it incredibly timely and useful.  Also of note:

Pawley's "Handbook of Biological Confocal Microscopy"
Periasamy's "Methods in Cellular Imaging"
Periasamy and Day, "Molecular Imaging, FRET Microscopy and Spectroscopy"
