Dear all,

I have an issue with ZDC unit (the old version ZDC, i.e. ZDC 1.) in our
IX81 TIRF microscope. I used it with MicroManager and it seems working.

I could manually find the focus and then find the offset, it looks working.
However when I turn on the autofocus, it is moving focus and defocusing
about 3um.
I tried to manually change the offset, but it does not take any effect.
The autofocus will focus ~3um away from the real focus.

Any suggestions? Thank you very much.

Dear Hongtao Chen,

Sounds like you did not implemented the offset and it finds the bottom of your sample. Did you have a look at: https://micro-manager.org/wiki/Olympus, last few lines.

Thanks for reply.

Well, I did actually. I do have the searching range 500 and use
"measureoffset now"  to find the offset, it usually gives 2.xx
Then when I turned on the autofocus. It will defocus and the zdc actually
find focus 3um above the real focus plane I need.
I then tried to manually play the offset (plus, minus, larger, smaller
numbers),
it does not take any effect on "autofocus on", still the same 3um above.
